Product Overview
This rabbit polyclonal antibody detects histone deacetylase 2 (gene HDAC2) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 55.4 kDa, and the supplier reports an observed band at about 60 kDa.
Histone deacetylase 2 (HDAC2) is a class I deacetylase closely related to HDAC1, and the two often act together in the Sin3, NuRD and CoREST corepressor complexes. Removing acetyl groups from histone tails generally compacts chromatin and represses transcription. HDAC2 is studied in gene silencing, memory formation and cancer, and it is a target of many HDAC inhibitors.

Samples with a confirmed band (WB): human K562 cells, human THP-1 cells, human HL-60 cells, human Caco-2 cells, human HepG2 cells, rat brain tissue, rat lung tissue, rat pancreas tissue, mouse brain tissue, mouse lung tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.25 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human breast cancer tissue, human colon cancer tissue, human thyroid carcinoma tissue, human laryngeal squamous cells carcinoma tissue, human ovarian serous tumor tissue, human lung squamous carcinoma tissue, mouse brain tissue, rat brain tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): Caco-2 cells.
Samples with a confirmed signal (flow cytometry): HL-60 cells.

Recombinant human HDAC2 fragment (Glu387–Pro488), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places histone deacetylase 2 in the nucleus and cytoplasm. Tissue expression noted by UniProt (human): Widely expressed. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
