Product Overview
This rabbit polyclonal antibody recognizes histone deacetylase 5 (gene HDAC5) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 122.0 kDa, and the supplier reports an observed band at about 150 kDa.
Histone deacetylase 5 (HDAC5) is a class IIa deacetylase that represses MEF2-dependent transcription while in the nucleus and is exported to the cytoplasm after phosphorylation. It helps control muscle differentiation and cardiac hypertrophy responses. HDAC5 is studied in cardiac remodeling, muscle biology and neuronal gene regulation.

Samples with a confirmed band (WB): human K562 cells, human PC-3 cells, rat liver tissue, mouse liver tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): mouse small intestine tissue, human placenta tissue, human esophageal squamous carcinoma tissue, human lung cancer tissue, human rectal cancer tissue, human breast cancer tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): A431 cells, U2OS cells.
Samples with a confirmed signal (flow cytometry): THP-1 cells.

Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places histone deacetylase 5 in the nucleus and cytoplasm. Tissue expression noted by UniProt (human): Widely expressed. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
