Product Overview
This rabbit polyclonal antibody recognizes histone deacetylase 8 (gene HDAC8) in human, mouse and rat samples and is validated for Western blot and ELISA. The predicted molecular weight is 41.8 kDa, and the supplier reports an observed band at about 50 kDa.
Histone deacetylase 8 (HDAC8) is a class I deacetylase that can act on histones, and its best-established substrate is the cohesin subunit SMC3. Deacetylating SMC3 recycles cohesin after cell division. HDAC8 mutations cause a form of Cornelia de Lange syndrome, and it is studied in cohesin biology and cancer.

Samples with a confirmed band (WB): rat heart tissue, rat brain tissue, mouse heart tissue, mouse brain tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Recombinant human HDAC8 fragment (Asp29–Glu347), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places histone deacetylase 8 in the nucleus, chromosome and cytoplasm. Tissue expression noted by UniProt (human): Weakly expressed in most tissues. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
