Product Overview
This rabbit polyclonal antibody recognizes WAP four-disulfide core domain protein 2 (gene WFDC2) in human, mouse and rat samples and is validated for ELISA, IHC and Western blot. The predicted molecular weight is 13.0 kDa, and the supplier reports an observed band at about 15 kDa.
WFDC2, better known as HE4 (human epididymis protein 4), is a small secreted protein carrying WAP four-disulfide core domains of the type found in protease inhibitors. It is expressed in epithelia of the reproductive tract and airways, animal studies show it is needed for normal lung and airway epithelial development, and serum HE4 is widely measured in ovarian cancer research.

Validated Applications
| Western blot | 0.1–0.5 µg/mL |
|---|---|
| IHC (paraffin sections) | 0.5–1 µg/mL |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): human HeLa cells, human MDA-MB-231 cells, human MDA-MB-453 cells, rat thymus tissue, mouse testis tissue, mouse thymus tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL, HRP-conjugated secondary antibody at 1:10000, ECL detection.
Samples with confirmed staining (IHC): rat small intestine tissue, mouse spleen tissue, rat spleen tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Immunogen
Recombinant human HE4 fragment (Glu31–Phe124), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt describes WAP four-disulfide core domain protein 2 as a secreted protein. Tissue expression noted by UniProt (human): Expressed in lung, trachea, kidney, breast, fallopian tubes, endometrium, cervix, epididymis and prostate. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
