Product Overview
This rabbit polyclonal antibody detects lymphoid-specific helicase (gene HELLS) in human and mouse samples and is validated for Western blot, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 97.1 kDa, and the supplier reports an observed band at about 97 kDa.
Lymphoid-specific helicase (HELLS, LSH) is an ATP-dependent chromatin remodeler of the SNF2 family. It helps DNA methyltransferases access nucleosomal DNA, supporting DNA methylation at repetitive sequences, heterochromatin formation and silencing of transposable elements, and it also aids DNA repair. HELLS mutations cause ICF syndrome, and it is studied in epigenetics and genome stability.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human, Mouse) |
|---|---|
| Immunocytochemistry / Immunofluorescence | 5 µg/mL (Human) |
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human) |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): human HeLa cells, human K562 cells, human PC-3 cells, mouse thymus tissue, mouse testis tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (ICC/IF): HeLa cells.
Samples with a confirmed signal (flow cytometry): PC-3 cells.
Immunogen
Recombinant human HELLS fragment (Asn167–Phe838), expressed in E. coli. Sequence identity with the mouse ortholog: 93%.
Reactivity Notes
The supplier lists reactivity with human and mouse. UniProt places lymphoid-specific helicase in the nucleus and chromosome. Tissue expression noted by UniProt (human): Highly expressed in proliferative tissues such as adult thymus and testis, and expressed at lower levels in uterus, small intestine, colon, and peripheral blood mononuclear cells. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
