Product Overview
This rabbit polyclonal antibody detects heme oxygenase 1 (gene Hmox1) in mouse and rat samples and is validated for Western blot and IHC. The predicted molecular weight is 32.9 kDa, and the supplier reports an observed band at about 33 kDa.
Heme oxygenase 1 (HO-1, HMOX1, also called HSP32) breaks down heme into biliverdin, ferrous iron and carbon monoxide. It is strongly induced by oxidative stress, heme and inflammation, and its products have antioxidant and anti-inflammatory effects, so HO-1 is a widely used marker of cellular stress responses and cytoprotection.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Mouse, Rat) |
|---|---|
| IHC (paraffin sections) | 2–5 µg/mL (Mouse, Rat) |
Samples with a confirmed band (WB): rat spleen tissue, rat liver tissue, rat PC-12 cells, mouse spleen tissue, mouse NIH/3T3 cells, mouse RAW264.7 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): mouse liver tissue, rat alcoholic hepatitis tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Immunogen
Recombinant mouse HMOX1 fragment (Glu2–Thr261), expressed in E. coli. Sequence identity with the human and rat orthologs: 82% and 93%, respectively.
Reactivity Notes
The supplier lists reactivity with mouse and rat. UniProt places heme oxygenase 1 in the endoplasmic reticulum membrane. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
