Product Overview
This rabbit polyclonal antibody detects small RNA 2'-O-methyltransferase (gene HENMT1) in human and mouse samples and is validated for Western blot, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 44.5 kDa, and the supplier reports an observed band at about 45 kDa.
HENMT1 (HEN1 methyltransferase homolog 1) is a small RNA methyltransferase that places a 2′-O-methyl mark on the 3′-terminal nucleotide of PIWI-interacting RNAs (piRNAs). This modification protects piRNAs from tailing and degradation, which is needed for piRNA-mediated transposon silencing in germ cells. HENMT1 is studied in germline development and small RNA biology.

Samples with a confirmed band (WB): human Jurkat cells, human MCF-7 cells, human HEL cells, mouse testis tissue, mouse RAW264.7 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (ICC/IF): HeLa cells.
Samples with a confirmed signal (flow cytometry): Jurkat cells.
Immunogen
Recombinant human HENMT1 fragment (Gln45–Asp388), expressed in E. coli. Sequence identity with the mouse and rat orthologs: 70.3% and 73.3%, respectively.
Reactivity Notes
The supplier lists reactivity with human and mouse. UniProt places small RNA 2'-O-methyltransferase in the cytoplasm. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
