Product Overview
This rabbit polyclonal antibody targets E3 ISG15--protein ligase HERC5 in human samples and is validated for Western blot, ICC/IF and ELISA. The predicted molecular weight is 116.9 kDa, and the supplier reports an observed band at about 117 kDa.
HERC5 is an interferon-induced HECT-type E3 ligase and the main enzyme that attaches the ubiquitin-like protein ISG15 to target proteins. Associated with polyribosomes, it modifies many newly made proteins, including viral proteins, and so supports innate antiviral defense. HERC5 is studied in interferon signaling and viral infection.

Samples with a confirmed band (WB): human 293T cells, human A549 cells, human PC-3 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (ICC/IF): U2OS cells.

Recombinant human HERC5 fragment (Phe793–Gly1024), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human. UniProt places E3 ISG15--protein ligase HERC5 in the cytoplasm. Tissue expression noted by UniProt (human): Expressed in testis and to a lesser degree in brain, ovary and placenta. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
