Product Overview
Clone FBB-8 is a rabbit monoclonal antibody that recognizes transcription factor HES-1 (gene HES1) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF and flow cytometry. The predicted molecular weight is 29.5 kDa, and the supplier reports an observed band at about 30 kDa.
HES1 is a basic helix-loop-helix transcriptional repressor and a primary target of Notch signaling. It binds N-box sites and represses proneural and differentiation genes, keeping progenitor and stem cells undifferentiated, and its expression oscillates in many cells. HES1 is widely studied in neural development, stem cell maintenance and cancer.

Samples with a confirmed band (WB): human U-87MG cells, human U251 cells, human 293T cells, rat brain tissue, mouse brain tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:500, ECL detection.
Samples with confirmed staining (IHC): human liver tissue, human breast cancer tissue, rat brain tissue, rat cerebellum tissue, rat kidney tissue, human thyriod cancer tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): rat kidney tissue, human breast cancer tissue, mouse brain tissue, mouse cerebellum tissue, human pancreas cancer tissue.
Immunogen
Synthetic peptide based on the sequence of human hes1.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places transcription factor HES-1 in the nucleus. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
