Product Overview
This rabbit polyclonal antibody detects beta-hexosaminidase subunit alpha (gene HEXA) in human, mouse and rat samples and is validated for Western blot, ICC/IF and ELISA. The predicted molecular weight is 60.7 kDa, and the supplier reports an observed band at about 60 kDa.
Beta-hexosaminidase subunit alpha (HEXA) is a lysosomal enzyme subunit that forms hexosaminidase A with the beta subunit (HEXB). Together with the GM2 activator protein, hexosaminidase A removes terminal N-acetylgalactosamine from GM2 ganglioside. HEXA mutations cause Tay–Sachs disease, and it is studied in lysosomal storage disorders.

Validated Applications
| Western blot | 0.1–0.25 µg/mL (Human, Mouse, Rat) |
|---|---|
| Immunocytochemistry / Immunofluorescence | 5 µg/mL (Human) |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): human 293T cells, human Caco-2 cells, human HeLa cells, rat kidney tissue, mouse kidney tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.25 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (ICC/IF): U2OS cells.
Immunogen
Recombinant human HEXA fragment (Ala135–His179), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places beta-hexosaminidase subunit alpha in the lysosome. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
