Product Overview
This rabbit polyclonal antibody targets hexokinase-2 (gene HK2) in human, mouse and rat samples and is validated for flow cytometry, IHC and Western blot. The predicted molecular weight is 102.4 kDa, and the supplier reports an observed band at about 102 kDa.
Hexokinase 2 (HK2) phosphorylates glucose to glucose 6-phosphate, the first committed step of glycolysis, and is the main hexokinase in insulin-sensitive tissues such as muscle and fat. It binds the outer mitochondrial membrane, where it helps protect cells from apoptosis, and its high expression in many tumors makes it a focus of cancer metabolism research.

Samples with a confirmed band (WB): human HeLa cells, human MCF-7 cells, human HepG2 cells, human HeLa- WT cells, human Hela-HK2 KO cells, rat skeletal muscle tissue, rat heart tissue, mouse skeletal muscle tissue, mouse heart tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 8% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human breast cancer tissue, mouse skeletal muscle tissue, rat skeletal muscle tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with a confirmed signal (flow cytometry): HepG2 cells.

Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places hexokinase-2 in the mitochondrion outer membrane and cytoplasm. Tissue expression noted by UniProt (human): Predominant hexokinase isozyme expressed in insulin-responsive tissues such as skeletal muscle. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
