Product Overview
Clone 17H18 is a rabbit monoclonal antibody that recognizes hypoxia-inducible factor 1-alpha (gene HIF1A) in human samples and is validated for Western blot, IHC, ICC/IF, immunoprecipitation and flow cytometry. The predicted molecular weight is 92.7 kDa, and the supplier reports an observed band at about 120 kDa.
HIF-1α (HIF1A) is the oxygen-regulated subunit of hypoxia-inducible factor 1. In normal oxygen it is hydroxylated and rapidly degraded, but under hypoxia it accumulates, pairs with ARNT (HIF-1β) and switches on genes for erythropoietin, VEGF, glucose uptake and glycolysis. It is a central marker in studies of hypoxia, angiogenesis, tumor metabolism and ischemia.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:1000, ECL detection.
Samples with confirmed staining (IHC): human kindey tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Samples with confirmed staining (ICC/IF): HeLa cells.
Immunogen
Synthetic peptide based on the sequence of human HIF-1 alpha.
Reactivity Notes
The supplier lists reactivity with human. UniProt places hypoxia-inducible factor 1-alpha in the cytoplasm, nucleus and nucleus speckle. Tissue expression noted by UniProt (human): Expressed in most tissues with highest levels in kidney and heart. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
