Product Overview
Clone HAD-8 is a rabbit monoclonal antibody that recognizes histone H2B type 1-K (gene H2BC12) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF, immunoprecipitation and flow cytometry. The predicted molecular weight is 13.9 kDa, and the supplier reports an observed band at about 17 kDa.
Histone H2B type 1-K (H2BC12) is a canonical H2B, one of the four core histones that make up the nucleosome. Nucleosomes package DNA into chromatin and control its access to transcription, replication and repair machinery, with post-translational histone marks adding a further layer of regulation. H2B ubiquitination and acetylation are well-known marks of active transcription.

Samples with a confirmed band (WB): human A431 cells, human SIHA cells, rat brain tissue, rat liver tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 12% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human lung cancer tissue, rat uterus tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with a confirmed result (IP): K562 cells.
Immunogen
Synthetic peptide based on the sequence of human histone H2B.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places histone H2B type 1-K in the nucleus and chromosome. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
