Product Overview

This rabbit polyclonal antibody targets major histocompatibility complex, class I, C (gene HLA-C) in human samples and is validated for flow cytometry, ICC/IF, IHC and Western blot.

HLA-C is a classical MHC class I molecule that presents intracellular peptides to CD8+ T cells. It is also a main ligand for killer-cell immunoglobulin-like receptors (KIRs) on natural killer cells, and it is studied in transplantation, viral immunity and pregnancy.

Target Major histocompatibility complex, class I, C (gene HLA-C; UniProt Q5QT33, human)
Host / Clonality / Isotype Rabbit / Polyclonal / Rabbit IgG
Reactivity Human
Form Lyophilized
Formulation Per vial: 0.9 mg NaCl; 0.2 mg Na2HPO4; 0.05 mg NaN3; plus stabilizers

Validated Applications

Western blot 0.1–0.5 µg/mL (Human)
IHC (paraffin sections) 0.5–1 µg/mL (Human)
Immunocytochemistry / Immunofluorescence 0.5–1 µg/mL (Human)
Immunocytochemistry / Immunofluorescence 2 µg/mL (Human)

Samples with a confirmed band (WB): human A431 cells, human 293T cells, human Jurkat cells, human U2OS cells, human HeLa cells, human HepG2 cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Samples with confirmed staining (IHC): human lung cancer tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Conditions in the example images (IHC): heat-mediated antigen retrieval in citrate buffer (pH 6).

Samples with confirmed staining (ICC/IF): HeLa cells.

Samples with confirmed staining (ICC/IF): human lung cancer tissue, human oesophagus squama cancer tissue.

Samples with a confirmed signal (flow cytometry): SiHa cells.

Immunogen

Synthetic peptide from the C-terminal region of human HLA-C.

Reactivity Notes

The supplier lists reactivity with human. UniProt places major histocompatibility complex, class I, C in the cell membrane and membrane. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.1–0.5 µg/mL (Human); IHC (paraffin sections) 0.5–1 µg/mL (Human); Immunocytochemistry / Immunofluorescence 2 µg/mL (Human); Flow cytometry (fixed cells) 1–3 µg/1×10⁶ cells (Human). Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human. Other species are not listed by the supplier.
Q.Has this antibody been tested on monkey samples?
A.Reactivity with monkey samples has not been tested by the supplier. Aligning the immunogen sequence with the monkey ortholog (e.g., by BLAST) helps estimate the chance of cross-reactivity.
Q.Is this antibody specific to HLA-C only?
A.According to the supplier: no. This antibody also cross-reacts with other HLA classes, such as HLA-B.
Q.Is a blocking reagent available for this antibody?
A.A matching blocking reagent for competition (specificity) controls can be supplied on request. Contact info@biohippo.com to request it.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: observed band at about 41 kDa.
