Product Overview
Clone 18H23 is a rabbit monoclonal antibody that recognizes high mobility group protein HMG-I/HMG-Y (gene HMGA1) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF and flow cytometry. The predicted molecular weight is 11.7 kDa, and the supplier reports an observed band at about 17 kDa.
HMGA1 (HMG-I/HMG-Y) is a small architectural chromatin protein that uses AT-hook motifs to bind the minor groove of AT-rich DNA. By bending DNA and helping assemble transcription factor complexes it tunes the activity of many genes. It is abundant in embryonic cells and many tumors, so it is studied in development and cancer.

Samples with a confirmed band (WB): human HepG2 cells, human A431 cells, human K562 cells, human PC-3 cells, rat C6 cells, mouse RAW264.7 cells, mouse NIH/3T3 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 12% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:1000 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human breast cancer tissue, human lung cancer tissue, human esophageal cancer tissue, human rectal cancer tissue, human ovarian cancer tissue, human liver cancer tissue, human pancreas cancer tissue, human placenta tissue, human stomach cancer tissue, mouse brain tissue, rat brain tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Immunogen
Synthetic peptide based on the sequence of human HMGA1.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places high mobility group protein HMG-I/HMG-Y in the nucleus and chromosome. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
