Product Overview
Clone 4F3 is a mouse monoclonal antibody that recognizes heterogeneous nuclear ribonucleoprotein D0 (gene HNRNPD) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF and flow cytometry. The predicted molecular weight is 38.4 kDa, and the supplier reports an observed band at about 43–45 kDa.
hnRNP D (HNRNPD, also called AUF1) binds AU-rich elements in the 3′ untranslated regions of many short-lived mRNAs, such as those of cytokines and proto-oncogenes, and usually promotes their decay. It also binds telomeric single-stranded DNA and regulates translation of some clock gene mRNAs, and it is studied in mRNA turnover and inflammation.

Samples with a confirmed band (WB): human HEK293 cells, human K562 cells, human A431 cells, human HepG2 cells, human placenta tissue, rat brain tissue, mouse brain tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:10000, ECL detection.
Samples with confirmed staining (IHC): human rectal cancer tissue, rat brain tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): A431 cells.
Samples with confirmed staining (ICC/IF): human rectal cancer tissue.
Samples with a confirmed signal (flow cytometry): SiHa cells, U937 cells.
Immunogen
Recombinant human hnRNP D/AUF1/HNRNPD fragment (Glu88–Asn246), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places heterogeneous nuclear ribonucleoprotein D0 in the nucleus and cytoplasm. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
