Product Overview
This rabbit polyclonal antibody targets protein Hook homolog 3 (gene HOOK3) in human, mouse and rat samples and is validated for ELISA, flow cytometry, ICC/IF, IHC and Western blot. The predicted molecular weight is 83.1 kDa, and the supplier reports an observed band at about 83 kDa.
Hook 3 (HOOK3) is a cargo adaptor for the microtubule motor dynein. Together with dynactin it switches dynein into a highly processive motor and links it to cargoes such as the FHF complex and endosomes. It also associates with the Golgi and centrosome, and it is studied in intracellular transport and neuronal development.

Samples with a confirmed band (WB): human MOLT-4 cells, human RT4 cells, human SiHa cells, rat brain tissue, mouse brain tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human placenta tissue, mouse testis tissue, human glioma tissue, human oesophagus squama cancer tissue, human sarcoma tissue, rat testis tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in citrate buffer (pH 6).
Samples with confirmed staining (ICC/IF): A431 cells.
Samples with a confirmed signal (flow cytometry): U87 cells.
Immunogen
Recombinant human HOOK3 fragment (Glu14–Arg198), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places protein Hook homolog 3 in the cytoplasm and Golgi apparatus. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
