Product Overview
Clone ADAB-8 is a rabbit monoclonal antibody that recognizes hypoxanthine-guanine phosphoribosyltransferase (gene HPRT1) in human, mouse and rat samples and is validated for Western blot, IHC and immunoprecipitation. The predicted molecular weight is 24.6 kDa, and the supplier reports an observed band at about 25 kDa.
Hypoxanthine-guanine phosphoribosyltransferase (HPRT1) is the key enzyme of the purine salvage pathway, recycling hypoxanthine and guanine into IMP and GMP. Complete loss of HPRT1 causes Lesch-Nyhan syndrome, and because it is steadily expressed it is also a common reference gene in expression studies.

Samples with a confirmed band (WB): human A549- WT cells, human A549-HPRT1 KO cells, human 293T cells, human U251 cells, rat brain tissue, rat C6 cells, mouse brain tissue, mouse Neuro-2a cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 12% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

The supplier lists reactivity with human, mouse and rat. UniProt places hypoxanthine-guanine phosphoribosyltransferase in the cytoplasm. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
