Biological background

Protein function (datasheet): Involved in stress resistance and actin organization.

Scientific background (datasheet): The heat-shock proteins (HSPs) belong to a larger group of polypeptides, the stress proteins, that are induced in various combinations in response to environmental challenges and developmental transitions. Heat-shock 27-kD protein1also knows as HSPB. Synthesis of the small (27-kD) HSP has been shown to be correlated with the acquisition of thermotolerance. HSP27 gene is mapped to 7q11.23. Mutant small heat-shock protein 27 causes axonal Charcot-Marie-Tooth disease and distal hereditary motor neuropathy. Heat shock protein 27 prevents cellular polyglutamine toxicity and suppresses the increase of reactive oxygen species caused by huntingtin.

Cellular localization (datasheet): Cytoplasm . Nucleus . Cytoplasm, cytoskeleton, spindle . Cytoplasmic in interphase cells. Colocalizes with mitotic spindles in mitotic cells. Translocates to the nucleus during heat shock and resides in sub-nuclear structures known as SC35 speckles or nuclear splicing speckles (By similarity). .

Tissue details (datasheet): Expressed in a variety of tissues. High levels in lung, adrenal, xiphoid, adipose tissue, heart and striated and smooth muscle, lower levels in the CNS. Adult levels are much higher in the slow-twitch soleus muscle than in the fast-twitch rectus femoris and extensor digitorum muscles. .

Sequence similarities (datasheet): Belongs to the small heat shock protein (HSP20) family.

Research relevance and current trends

  • Commonly studied in contexts related to Atherosclerosis,Cancer,Cardiovascular,Chaperones,Contractility,Heat Shock Proteins,Protein Trafficking,Signal Transduction,Tumor Biomarkers.
  • Supports comparative expression analysis across conditions, genotypes, or treatments when paired with appropriate controls.
  • Useful for confirming target presence and subcellular distribution using orthogonal readouts (e.g., microscopy vs. immunoblotting).

Common research applications

  • Western blot (WB): Compare relative target abundance and apparent size/isoforms across samples; interpret bands in light of expected MW and potential PTMs.
  • Immunohistochemistry (IHC): Assess tissue distribution and cell-type patterns; interpret staining with appropriate negative controls and antigen context.
  • Immunofluorescence / ICC: Visualize subcellular localization and co-localization patterns; consider fixation/permeabilization compatibility and controls.

Notes for experimental interpretation

  • Consider isoforms, post-translational modifications, and processing that can shift apparent molecular weight or localization.
  • Cross-reactivity (datasheet): No cross-reactivity with other proteins
  • Use appropriate positive and negative controls (e.g., KO/KD, blocking peptide, or isotype controls) to support specificity interpretation.

As a monoclonal antibody, this reagent is expected to recognize a defined epitope, which can support consistency across lots when epitope accessibility is preserved.
