Product Overview
Clone HII-8 is a rabbit monoclonal antibody that recognizes heat shock protein beta-1 (gene HSPB1) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF, immunoprecipitation and flow cytometry. The predicted molecular weight is 22.8 kDa, and the supplier reports an observed band at about 27 kDa.
HSP27 (HSPB1) is a small heat shock protein that forms large oligomers and binds unfolding proteins to prevent their aggregation. Its phosphorylation by p38 MAPK-activated kinases regulates its assembly and roles in actin organization and resistance to apoptosis, and it is a common marker of cellular stress. HSPB1 mutations cause inherited peripheral neuropathy.

Samples with a confirmed band (WB): human HeLa cells, human MCF-7 cells, human 293T cells, human K562 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:500, ECL detection.
Samples with confirmed staining (IHC): human colon cancer tissue, rat stomach tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Immunogen
Synthetic peptide based on the sequence of human hsp27.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places heat shock protein beta-1 in the cytoplasm and nucleus. Tissue expression noted by UniProt (human): Detected in all tissues tested: skeletal muscle, heart, aorta, large intestine, small intestine, stomach, esophagus, bladder, adrenal gland, thyroid, pancreas, testis, adipose tissue, kidney, liver, spleen, cerebral cortex, blood serum and cerebrospinal fluid. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
