Product Overview

Clone DOI-8 is a rabbit monoclonal antibody that recognizes 60 kDa heat shock protein, mitochondrial (gene HSPD1) in human, mouse and rat samples and is validated for Western blot and IHC. The predicted molecular weight is 61.1 kDa, and the supplier reports an observed band at about 60 kDa.

HSP60 (HSPD1) is the mitochondrial chaperonin. It forms barrel-shaped complexes that, together with the lid-like co-chaperonin HSP10, enclose imported proteins and help them fold in the mitochondrial matrix. HSP60 is studied in mitochondrial protein quality control, and it is also known as an immune target in inflammation research.

Validated Applications

Western blot 1:500–1:2000
Immunohistochemistry 1:50–1:200

Samples with confirmed staining (IHC): human kidney tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Immunogen

Synthetic peptide based on the sequence of human hsp60.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places 60 kDa heat shock protein, mitochondrial in the mitochondrion matrix. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000; Immunohistochemistry 1:50–1:200. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 61.1 kDa; observed band at about 60 kDa.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human hsp60.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
