Rabbit Monoclonal

Product Overview

Clone 19I70 is a rabbit monoclonal antibody that recognizes immunoglobulin heavy constant gamma 2 (gene IGHG2) in human samples and is validated for Western blot, IHC and ICC/IF. The predicted molecular weight is 35.9 kDa, and the supplier reports an observed band at about 53 kDa.

IGHG2 encodes the constant region of the immunoglobulin γ2 heavy chain, which defines the IgG2 subclass. IgG2 dominates antibody responses to bacterial capsular polysaccharides and has relatively weak interactions with Fcγ receptors and complement compared with IgG1 and IgG3.

Validated Applications

Western blot 1:1000–1:5000
Immunohistochemistry 1:50–1:200
Immunocytochemistry / Immunofluorescence 1:50–1:200

Samples with a confirmed band (WB): human spleen.

Recommended loading (WB): 20–40 µg of total protein per lane.

Samples with confirmed staining (IHC): human tonsil tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).

Immunogen

Synthetic peptide based on the sequence of human igG2.

Reactivity Notes

The supplier lists reactivity with human. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:1000–1:5000; Immunohistochemistry 1:50–1:200; Immunocytochemistry / Immunofluorescence 1:50–1:200. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 35.9 kDa; observed band at about 53 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human igG2.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
