Immunogen A synthesized peptide derived from human Human IgG3 IgG is a monomeric immunoglobulin, built of two heavy chains gamma and two light chains. Each molecule has two antigen binding sites. This is the most abundant immunoglobulin and is approximately equally distributed in blood and in tissue liquids, constituting 75% of serum immunoglobulins in humans. There are 4 subclasses: IgG1 (66%), IgG2 (23%), IgG3 (7%) and IgG4 (4%) .

Product Overview

Clone ACIA-9 is a rabbit monoclonal antibody that recognizes immunoglobulin heavy constant gamma 3 (gene IGHG3) in human samples and is validated for Western blot and immunoprecipitation. The predicted molecular weight is 41.3 kDa, and the supplier reports an observed band at about 55 kDa.

IGHG3 encodes the constant region of the immunoglobulin γ3 heavy chain, which defines IgG3. This subclass has an unusually long hinge region, strongly activates complement and Fcγ receptors, and has a shorter serum half-life than other IgG subclasses.

Validated Applications

Western blot 1:500–1:2000
Immunoprecipitation 1:50

Samples with a confirmed band (WB): human plasma.

Recommended loading (WB): 20–40 µg of total protein per lane.

Immunogen

Synthetic peptide based on the sequence of human human IgG3 IgG is a monomeric immunoglobulin, built of two heavy chains gamma and two light chains.

Reactivity Notes

The supplier lists reactivity with human. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000; Immunoprecipitation 1:50. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human. Other species are not listed by the supplier.
Q.Is a carrier-free (BSA-free) version available, for example for conjugation?
A.Yes. A carrier-free formulation without BSA, stabilized with trehalose and/or glycerol, can be prepared on request and is compatible with common conjugation chemistries. Please note this when ordering or contact info@biohippo.com. For storage at −20 °C, keep a cryoprotectant such as glycerol in the buffer rather than PBS alone.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 41.3 kDa; observed band at about 55 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human human IgG3 IgG is a monomeric immunoglobulin, built of two heavy chains gamma and two light chains.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
