Product Overview
This rabbit polyclonal antibody recognizes intercellular adhesion molecule 1 (gene Icam1) in mouse samples and is validated for Western blot, IHC and ELISA. The predicted molecular weight is 58.8 kDa, and the supplier reports an observed band at about 115 kDa.
ICAM-1 (CD54) is a cell surface glycoprotein of the immunoglobulin superfamily that is strongly induced on endothelial and immune cells by inflammatory cytokines. It binds the integrins LFA-1 and Mac-1 on leukocytes, supporting their firm adhesion and passage across blood vessel walls as well as T-cell activation, and it is a widely used marker of endothelial activation and inflammation.

Validated Applications
| Western blot | 0.1–0.25 µg/mL (Mouse) |
|---|---|
| IHC (paraffin sections) | 2–5 µg/mL (Mouse) |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): mouse spleen tissue, mouse thymus tissue, mouse liver tissue, mouse kidney tissue, mouse lung tissue, mouse RAW264.7 cells, mouse ANA-1 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.25 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): mouse intestine tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Immunogen
Recombinant mouse Icam1 fragment (Lys53–Pro537), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with mouse. UniProt places intercellular adhesion molecule 1 in the cell membrane. Tissue expression noted by UniProt (mouse): Expressed at low level on a subpopulation of lymphocytes, macrophages, and endothelial cells, but is strongly induced on these cells, and on fibroblasts and epithelial cells. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
