Product Overview
This rabbit polyclonal antibody detects protein-S-isoprenylcysteine O-methyltransferase (gene ICMT) in human and rat samples and is validated for Western blot and ELISA. The predicted molecular weight is 31.9 kDa, and the supplier reports an observed band at about 32 kDa.
ICMT (isoprenylcysteine carboxyl methyltransferase) is an enzyme of the endoplasmic reticulum that methylates the C-terminal cysteine of prenylated proteins after the CAAX processing steps. This final modification affects the membrane targeting and function of proteins such as RAS and Rho family GTPases, so ICMT is studied in cell signaling and cancer biology.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Rat) |
|---|---|
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): rat brain tissue, rat C6 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Immunogen
Recombinant human ICMT fragment (Glu13–Leu284), expressed in E. coli. Sequence identity with the mouse and rat orthologs: 96.3% and 95.2%, respectively.
Reactivity Notes
The supplier lists reactivity with human and rat. UniProt places protein-S-isoprenylcysteine O-methyltransferase in the endoplasmic reticulum membrane. Tissue expression noted by UniProt (human): Ubiquitously expressed. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
