Product Overview
Clone 23I42 is a rabbit monoclonal antibody that recognizes inducible T-cell costimulator (gene ICOS) in human, mouse and rat samples and is validated for Western blot, ICC/IF and flow cytometry. The predicted molecular weight is 22.6 kDa, and the supplier reports an observed band at about 23 kDa.
ICOS (inducible T-cell costimulator, CD278) is a CD28-family receptor expressed on activated and memory T cells. When engaged by ICOS ligand on antigen-presenting cells, it boosts T-cell proliferation and cytokine production and is essential for T follicular helper cell development and T-cell help to B cells, making it a focus of germinal center and immunotherapy research.

|---|---|
| Immunocytochemistry / Immunofluorescence | 1:50–1:200 |
| Flow cytometry | 1:50 |
Samples with a confirmed band (WB): human Jurkat cells, human HEL cells, rat spleen tissue, mouse spleen tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:500, ECL detection.
Immunogen
Synthetic peptide based on the sequence of human ICOS.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. Tissue expression noted by UniProt (human): Activated T-cells. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
