Key elements and design rationale

  • Target: IDH2 (Glucocorticoid receptor).
  • Antibody format: Monoclonal; clone ACFO-9; isotype Rabbit IgG.
  • Host: Rabbit.
  • Species reactivity: Human,Mouse,Rat (confirm in your model system with appropriate controls).

This description is intended to help interpret the antibody design and the biological context of the target using the fields provided in the catalog record, alongside general experimental considerations.

Biological background

IDH2 (protein: T-cell surface glycoprotein CD4) is a commonly studied target in molecular and cellular biology. Functional context (as provided): Receptor for glucocorticoids (GC). Has a dual mode of action: as a transcription factor that binds to glucocorticoid response elements (GRE), both for nuclear and mitochondrial DNA, and as a modulator of other transcription factors. Affects inflammatory responses, cellular proliferation and differentiation in target tissues. Could act as a coactivator for STAT5-dependent transcription upon growth hormone (GH) stimulation and could reveal an essential role of hepatic GR in the control of body growth. Involved in chromatin remodeling. May play a negative role in adipogenesis through the regulation of lipolytic and antilipogenic genes expression. . Reported cellular localization context: Cytoplasm . Mitochondrion. Nucleus . Cytoplasmic in the absence of ligand, nuclear after ligand- binding. Tissue expression notes (as provided): Widely expressed. In the heart, detected in left and right atria, left and right ventricles, aorta, apex, intraventricular septum, and atrioventricular node as well as whole adult and fetal heart. .

Research relevance and current trends

  • Research context keywords from the source record include: Cancer,Cancer Metabolism,Energy Metabolism,Energy Transfer Pathways,Integration Of Energy,Integration Of Energy Metabolism,Metabolic Signaling Pathway,Metabolic Signaling Pathways,Metabolism,Mitochondrial,Mitochondrial Markers,Mitochondrial Metabolism,Pathways and Processes,Signal Transduction.
  • Current studies often focus on connecting target abundance/localization to pathway perturbations across models, tissues, and cell states.
  • Quantitative and multiplexed assays (e.g., imaging + immunoblot panels) are commonly used to compare phenotypes across conditions and time-courses.

Common research applications

  • Western blotting (WB): assess relative target abundance across samples, treatments, or time-points.
  • Immunohistochemistry (IHC): evaluate spatial distribution of target-positive staining in tissue architecture.
  • Flow cytometry: quantify target-positive populations and compare shifts in marker distributions.

Workflow ideas (metafield): Validate IDH2 antibody specificity using KO/KD control samples (WB/IF/IHC as appropriate), Detect IDH2 expression by Western blot in cell or tissue lysates, Detect IDH2 in FFPE tissue sections by immunohistochemistry, Quantify IDH2-positive cells by flow cytometry in single-cell suspensions

Notes for experimental interpretation

  • Consider isoforms and post-translational modifications (PTMs) that may shift apparent molecular weight or epitope accessibility.
  • Apparent molecular weight may vary by sample type and processing (observed MW: 30 kDa; calculated MW: 85659 MW).
  • Control concepts: include appropriate negative controls (e.g., isotype, KO/KD samples) and orthogonal validation when feasible.

Additional product details (from the source record)

  • Molecular weight (observed): 30 kDa
  • Cellular localization (provided): Cytoplasm . Mitochondrion. Nucleus . Cytoplasmic in the absence of ligand, nuclear after ligand- binding.
  • Tissue details (provided): Widely expressed. In the heart, detected in left and right atria, left and right ventricles, aorta, apex, intraventricular septum, and atrioventricular node as well as whole adult and fetal heart. .

We are currently using anti-IDH2 Monoclonal antibody M00510 for mouse tissue, and we are content with the WB results. The species of reactivity given in the datasheet says human, mouse, rat. Is it likely that the antibody can work on zebrafish tissues as well?
The anti-IDH2 Monoclonal antibody (M00510) has not been tested for cross reactivity specifically with zebrafish tissues, though there is a good chance of cross reactivity. We have an innovator award program that if you test this antibody and show it works in zebrafish you can get your next antibody for free. Please contact me if I can help you with anything.
I was wanting to use your anti-IDH2 Monoclonal antibody for WB for mouse colon skin on frozen tissues, but I want to know if it has been validated for this particular application. Has this antibody been validated and is this antibody a good choice for mouse colon skin identification?
As indicated on the product datasheet, M00510 anti-IDH2 Monoclonal antibody has been validated for WB on human, mouse, rat tissues. We have an innovator award program that if you test this antibody and show it works in mouse colon skin in IHC-frozen, you can get your next antibody for free.
