This description is intended to help interpret the antibody design and the biological context of the target using the fields provided in the catalog record, alongside general experimental considerations.

    Biological background

IFNAR1 (protein: P2X purinoceptor 1) is a commonly studied target in molecular and cellular biology. Functional context (as provided): Component of the receptor for type I interferons, including interferons alpha, IFNB1 and IFNW1 (PubMed:2153461, PubMed:7665574, PubMed:10049744, PubMed:14532120, PubMed:15337770, PubMed:21854986). Functions in general as heterodimer with IFNAR2 (PubMed:7665574, PubMed:10049744, PubMed:21854986). Type I interferon binding activates the JAK-STAT signaling cascade, and triggers tyrosine phosphorylation of a number of proteins including JAKs, TYK2, STAT proteins and the IFNR alpha- and beta- subunits themselves (PubMed:7665574, PubMed:21854986). Can form an active IFNB1 receptor by itself and activate a signaling cascade that does not involve activation of the JAK-STAT pathway (By similarity). . Reported cellular localization context: Isoform 1: Cell membrane ; Single- pass type I membrane protein . Late endosome . Lysosome . Interferon binding triggers internalization of the receptor from the cell membrane into endosomes and then into lysosomes. . Tissue expression notes (as provided): IFN receptors are present in all tissues and even on the surface of most IFN-resistant cells. Isoform 1, isoform 2 and isoform 3 are expressed in the IFN-alpha sensitive myeloma cell line U266B1. Isoform 2 and isoform 3 are expressed in the IFN-alpha resistant myeloma cell line U266R. Isoform 1 is not expressed in IFN-alpha resistant myeloma cell line U266R. .

    Research relevance and current trends

    • Research context keywords from the source record include: Cancer,Cytokines,Epigenetics and Nuclear Signaling,Immunology,Innate Immunity,Interferons,Nuclear Signaling,Nuclear Signaling Pathways,Signal Transduction,Signaling Pathway,Tumor Immunology.
    • Current studies often focus on connecting target abundance/localization to pathway perturbations across models, tissues, and cell states.
    • Quantitative and multiplexed assays (e.g., imaging + immunoblot panels) are commonly used to compare phenotypes across conditions and time-courses.

    Common research applications

    • Western blotting (WB): assess relative target abundance across samples, treatments, or time-points.
    • Flow cytometry: quantify target-positive populations and compare shifts in marker distributions.
    • Immunoprecipitation (IP): enrich target complexes for downstream immunoblot or interaction analyses.

Workflow ideas (metafield): Validate IFNAR1 antibody specificity using KO/KD control samples (WB/IF/IHC as appropriate), Detect IFNAR1 expression by Western blot in cell or tissue lysates, Quantify IFNAR1-positive cells by flow cytometry in single-cell suspensions, Enrich IFNAR1 by immunoprecipitation from lysates for downstream analysis

    Notes for experimental interpretation

    • Consider isoforms and post-translational modifications (PTMs) that may shift apparent molecular weight or epitope accessibility.
    • Apparent molecular weight may vary by sample type and processing (observed MW: 62 kDa; calculated MW: 63525 MW).
    • Control concepts: include appropriate negative controls (e.g., isotype, KO/KD samples) and orthogonal validation when feasible.

Additional product details (from the source record)

    • Molecular weight (observed): 62 kDa
    • Cellular localization (provided): Isoform 1: Cell membrane ; Single- pass type I membrane protein . Late endosome . Lysosome . Interferon binding triggers internalization of the receptor from the cell membrane into endosomes and then into lysosomes. .

We are currently using anti-IFNAR1/Beta R1 Rabbit Monoclonal antibody M00306-1 for human tissue, and we are content with the WB results. The species of reactivity given in the datasheet says human. Is it likely that the antibody can work on goat tissues as well?
The anti-IFNAR1/Beta R1 Rabbit Monoclonal antibody (M00306-1) has not been validated for cross reactivity specifically with goat tissues, though there is a good chance of cross reactivity. We have an innovator award program that if you test this antibody and show it works in goat you can get your next antibody for free. Please contact me if I can help you with anything.
Would M00306-1 anti-IFNAR1/Beta R1 Rabbit Monoclonal antibody work on parafin embedded sections? If so, which fixation method do you recommend we use (PFA, paraformaldehyde, other)?
It shows on the product datasheet, M00306-1 anti-IFNAR1/Beta R1 Rabbit Monoclonal antibody as been tested on WB. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.
