Product Overview
This rabbit polyclonal antibody recognizes intraflagellar transport protein 74 homolog (gene IFT74) in human, mouse, rat and monkey samples and is validated for Western blot, IHC, flow cytometry and ELISA. The predicted molecular weight is 69.2 kDa, and the supplier reports an observed band at about 69 kDa.
IFT74 is a component of intraflagellar transport complex B that, together with IFT81, forms a module that binds tubulin and carries it into the growing cilium. It is required for ciliogenesis and for building the sperm flagellum.

Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human esophageal squamous carcinoma tissue, human lung cancer tissue, human rectum adenocarcinoma tissue, mouse testis tissue, rat testis tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with a confirmed signal (flow cytometry): Jurkat cells.

Reactivity Notes
The supplier lists reactivity with human, mouse, rat and monkey. UniProt places intraflagellar transport protein 74 homolog in the cell projection and cytoplasmic vesicle. Tissue expression noted by UniProt (human): Highly expressed in adult and fetal kidney and expressed at lower level in adult heart, placenta, lung, liver and pancreas, and in fetal heart, lung and liver. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
