Product Overview

This rabbit polyclonal antibody detects insulin-like growth factor-binding protein 3 (gene IGFBP3) in human, mouse and rat samples and is validated for ELISA, IHC and Western blot. The predicted molecular weight is 31.7 kDa, and the supplier reports an observed band at about 40 kDa.

IGFBP-3 is the most abundant insulin-like growth factor binding protein in the circulation, where it forms a ternary complex with IGF-1 or IGF-2 and the acid-labile subunit that prolongs their half-life. It also has IGF-independent antiproliferative and pro-apoptotic effects, and its circulating levels are widely measured alongside IGF-1 in growth research.

Validated Applications

IHC (paraffin sections) 0.5–1 µg/mL (Human)
ELISA 0.1–0.5 µg/mL
Western blot 0.1–0.5 µg/mL (Human, Mouse, Rat)

Samples with a confirmed band (WB): human A431 cells, rat kidney tissue, rat liver tissue, mouse kidney tissue, mouse liver tissue.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Samples with confirmed staining (IHC): human intestinal cancer tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Conditions in the example images (IHC): heat-mediated antigen retrieval in citrate buffer (pH 6).

Immunogen

Synthetic peptide from the C-terminal region of human IGFBP-3. The peptide sequence is conserved in mouse and rat.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt describes insulin-like growth factor-binding protein 3 as secreted and also located in the nucleus. Tissue expression noted by UniProt (human): Expressed by most tissues. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.1–0.5 µg/mL (Human, Mouse, Rat); IHC (paraffin sections) 0.5–1 µg/mL (Human); ELISA 0.1–0.5 µg/mL. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 31.7 kDa; observed band at about 40 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide from the C-terminal region of human IGFBP-3. The peptide sequence is conserved in mouse and rat.
Q.How should this antibody be stored?
A.As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.
