Product Overview
Clone AOOA-9 is a rabbit monoclonal antibody that recognizes inhibitor of nuclear factor kappa-B kinase subunit beta (gene IKBKB) in human, mouse and rat samples and is validated for Western blot, IHC and ICC/IF. The predicted molecular weight is 86.6 kDa, and the supplier reports an observed band at about 87 kDa.
IKKβ (IKBKB) is the main catalytic subunit of the IKK complex in the canonical NF-κB pathway. In response to cytokines, microbial products and stress, it phosphorylates IκB proteins, targeting them for proteasomal degradation and releasing NF-κB to enter the nucleus, so it is a central kinase in inflammation research.

Samples with a confirmed band (WB): human HeLa cells, human Raji cells, human A549 cells, rat heart tissue, mouse heart tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human kidney tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Samples with confirmed staining (ICC/IF): HeLa cells.

Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places inhibitor of nuclear factor kappa-B kinase subunit beta in the cytoplasm, nucleus and membrane raft. Tissue expression noted by UniProt (human): Highly expressed in heart, placenta, skeletal muscle, kidney, pancreas, spleen, thymus, prostate, testis and peripheral blood. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
