This description is intended to help interpret the antibody design and the biological context of the target using the fields provided in the catalog record, alongside general experimental considerations.

Biological background

IKBKB (protein: P2X purinoceptor 1) is a commonly studied target in molecular and cellular biology. Functional context (as provided): Serine kinase that plays an essential role in the NF- kappa-B signaling pathway which is activated by multiple stimuli such as inflammatory cytokines, bacterial or viral products, DNA damages or other cellular stresses. Acts as part of the canonical IKK complex in the conventional pathway of NF-kappa-B activation and phosphorylates inhibitors of NF-kappa-B on 2 critical serine residues. These modifications allow polyubiquitination of the inhibitors and subsequent degradation by the proteasome. In turn, free NF-kappa-B is translocated into the nucleus and activates the transcription of hundreds of genes involved in immune response, growth control, or protection against apoptosis. In addition to the NF-kappa-B inhibitors, phosphorylates several other components of the signaling pathway including NEMO/IKBKG, NF-kappa-B subunits RELA and NFKB1, as well as IKK-related kinases TBK1 and IKBKE. IKK-related kinase phosphorylations may prevent the overproduction of inflammatory mediators since they exert a negative regulation on canonical IKKs. Also phosphorylates other substrates including NCOA3, BCL10 and IRS1. Within the nucleus, acts as an adapter protein for NFKBIA degradation in UV-induced NF-kappa-B activation. . Reported cellular localization context: Cytoplasm. Nucleus. Membrane raft. Colocalized with DPP4 in membrane rafts. Tissue expression notes (as provided): Highly expressed in heart, placenta, skeletal muscle, kidney, pancreas, spleen, thymus, prostate, testis and peripheral blood.

Research relevance and current trends

  • Research context keywords from the source record include: Innate Immunity,NFKB Pathway,Nuclear Signaling Pathways,Protein Phosphorylation,Ser/Thr Kinases,Signal Transduction,TLR Signaling,Vascular Inflammation.
  • Current studies often focus on connecting target abundance/localization to pathway perturbations across models, tissues, and cell states.
  • Quantitative and multiplexed assays (e.g., imaging + immunoblot panels) are commonly used to compare phenotypes across conditions and time-courses.

Common research applications

  • Western blotting (WB): assess relative target abundance across samples, treatments, or time-points.
  • Immunohistochemistry (IHC): evaluate spatial distribution of target-positive staining in tissue architecture.
  • Immunofluorescence/ICC (IF/ICC): visualize subcellular localization patterns and cell-to-cell heterogeneity.

Workflow ideas (metafield): Validate IKBKB antibody specificity using KO/KD control samples (WB/IF/IHC as appropriate), Detect IKBKB expression by Western blot in cell or tissue lysates, Detect IKBKB in FFPE tissue sections by immunohistochemistry, Localize IKBKB by immunofluorescence/immunocytochemistry in cultured cells

Notes for experimental interpretation

  • Consider isoforms and post-translational modifications (PTMs) that may shift apparent molecular weight or epitope accessibility.
  • Apparent molecular weight may vary by sample type and processing (observed MW: 87 kDa; calculated MW: 86564 MW).
  • Control concepts: include appropriate negative controls (e.g., isotype, KO/KD samples) and orthogonal validation when feasible.

Additional product details (from the source record)

  • Molecular weight (observed): 87 kDa
  • Cellular localization (provided): Cytoplasm. Nucleus. Membrane raft. Colocalized with DPP4 in membrane rafts.
  • Tissue details (provided): Highly expressed in heart, placenta, skeletal muscle, kidney, pancreas, spleen, thymus, prostate, testis and peripheral blood.
