Product Overview
This rabbit polyclonal antibody detects interleukin-2 receptor subunit alpha (gene Il2ra) in mouse and rat samples and is validated for Western blot, IHC and flow cytometry. The predicted molecular weight is 30.7 kDa, and the supplier reports an observed band at about 50 kDa.
IL-2 receptor alpha (CD25) is the subunit that gives the IL-2 receptor high affinity for IL-2. It is highly expressed on regulatory T cells and on activated T cells, and it is essential for Treg maintenance and immune tolerance. CD25 is a common marker for Tregs and T-cell activation, and its soluble form is measured in inflammation studies.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Mouse, Rat) |
|---|---|
| IHC (paraffin sections) | 2–5 µg/mL (Mouse, Rat) |
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Mouse) |
Samples with a confirmed band (WB): mouse ANA-1 cells, mouse NIH/3T3 cells, mouse RAW264.7 cells, mouse HEPA1–6 cells, rat C6 cells, rat NRK cells, rat RH35 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): mouse spleen tissue, rat small intestine tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with a confirmed signal (flow cytometry): HEPA1–6 cells.
Immunogen
Synthetic peptide from the middle region of mouse IL-2 Receptor alpha/Il2ra. Sequence identity with the human and rat orthologs: 64.3% and 86.7%, respectively.
Reactivity Notes
The supplier lists reactivity with mouse and rat. UniProt places interleukin-2 receptor subunit alpha in the membrane. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
