Product Overview
This rabbit polyclonal antibody detects interleukin-31 (gene IL31) in human, mouse and rat samples and is validated for Western blot, IHC and ELISA. The predicted molecular weight is 18.2 kDa, and the supplier reports an observed band at about 30–35 kDa.
Interleukin-31 (IL-31) is a cytokine produced mainly by activated Th2 cells. It signals through a receptor made of IL-31RA and OSMR, activating JAK1/JAK2 and STAT3/STAT5 as well as MAPK pathways. IL-31 is well known for its role in itch and skin inflammation, and it is studied in atopic dermatitis research.

Samples with a confirmed band (WB): human K562 cells, human SH-SY5Y cells, human Caco-2 cells, rat testis tissue, rat PC-12 cells, mouse testis tissue, mouse RAW264.7 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 12% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human tosil tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).

Recombinant human IL-31 fragment (Ser24–Thr164), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt describes interleukin-31 as a secreted protein. Tissue expression noted by UniProt (human): Detected at low levels in testis, bone marrow, skeletal muscle, kidney, colon, thymus, small intestine and trachea. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
