Overview

Anti-IL-31 Antibody Picoband® is an antibody for IL31 detection raised in Rabbit (Polyclonal, Rabbit IgG), with reported reactivity: Human,Mouse,Rat. Commonly used in WB, IHC, Flow Cytometry, ELISA workflows.

Key elements and design rationale

  • Target: IL31 (interleukin 31); UniProt: Q6EBC2
  • Antibody format: Rabbit, Polyclonal, Rabbit IgG
  • Molecular weight: 29 kDa
  • Applications: WB, IHC, Flow Cytometry, ELISA

Vendor description (summary): Boster Bio Anti-IL-31 Antibody Picoband® catalog # A08339-1.

Biological background

Biological context: Activates STAT3 and possibly STAT1 and STAT5 through the IL31 heterodimeric receptor composed of IL31RA and OSMR. May function in skin immunity. Enhances myeloid progenitor cell survival in vitro. Induces RETNLA and serum amyloid A protein expression in macrophages.

Expression and localization notes: cellular localization: Secreted., tissue context: Detected at low levels in testis, bone marrow, skeletal muscle, kidney, colon, thymus, small intestine and trachea..

Common research applications

  • Western blotting (WB): Compare IL31 levels across samples and conditions using appropriate loading and biological controls.
  • Immunohistochemistry (IHC): Evaluate spatial distribution of IL31 in tissue sections, considering fixation and antigen retrieval effects.
  • Flow cytometry: Quantify IL31-positive populations in single-cell suspensions with appropriate gating and controls.
  • ELISA: Use antibody-based detection formats to assess antigen presence or binding in plate-based assays.

Notes for experimental interpretation

  • Account for isoforms, post-translational modifications, and sample-specific processing that can shift apparent molecular weight or epitope accessibility.
  • Use positive/negative biological controls where possible (e.g., known-expressing cells/tissues, knockdown/knockout models) and include appropriate secondary-only/isotype controls for imaging workflows.

Additional product notes (from provided fields)

  • Specificity: No cross reactivity with other proteins.
  • Background: Interleukin-31 (IL-31) is a protein that in humans is encoded by the IL31 gene that resides on chromosome 12. IL31, which is made principally by activated Th2-type T cells, interacts with a heterodimeric receptor consisting of IL31RA and OSMR that is constitutively expressed on epithelial cells and keratinocytes. IL31 may be involved in the promotion of allergic skin disorders and in regulating other allergic diseases, such as asthma.
  • Cross reactivity: No cross-reactivity with other proteins.
  • Cellular localization: Secreted.
  • Tissue details: Detected at low levels in testis, bone marrow, skeletal muscle, kidney, colon, thymus, small intestine and trachea.
  • Research category: Immune System Diseases,Immunology,Protein Trafficking,Signal Transduction,Signaling Pathway

My colleagues were happy with the WB result of your anti-IL-31 antibody. However we have been able to see positive staining in testis secreted. using this antibody. Is that expected? Could you tell me where is IL31 supposed to be expressed?
From literature, testis does express IL31. Generally IL31 expresses in secreted. Regarding which tissues have IL31 expression, here are a few articles citing expression in various tissues: Peripheral blood, Pubmed ID: 15184896
I was wanting to use your anti-IL-31 antibody for IHC-P for mouse peripheral blood on frozen tissues, but I want to know if it has been validated for this particular application. Has this antibody been validated and is this antibody a good choice for mouse peripheral blood identification?
It shows on the product datasheet, A08339-1 anti-IL-31 antibody has been validated for ELISA, IHC-P, WB on human, mouse, rat tissues. We have an innovator award program that if you test this antibody and show it works in mouse peripheral blood in IHC-frozen, you can get your next antibody for free.
