Product Overview
This rabbit polyclonal antibody recognizes pro-interleukin-16 (gene IL16) in human samples and is validated for ELISA, IHC, ICC/IF and Western blot. The predicted molecular weight is 141.8 kDa, and the supplier reports an observed band at about 45–55 kDa.
Interleukin-16 (IL-16) is a cytokine released from a larger precursor after caspase-3 cleavage and acts as a chemoattractant for CD4+ cells, including T cells, monocytes and eosinophils. It uses CD4 as a receptor, and the pro-IL-16 precursor also has intracellular roles, including in T-cell cycle control.

Samples with a confirmed band (WB): human U937 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL, HRP-conjugated secondary antibody at 1:10000, ECL detection.
Samples with confirmed staining (IHC): human tonsil tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Samples with confirmed staining (ICC/IF): human cord blood.
Immunogen
Recombinant human IL-16 fragment (Ser1212–Ser1332), expressed in E. coli. Sequence identity with the mouse ortholog: 86%.
Reactivity Notes
The supplier lists reactivity with human. UniProt places pro-interleukin-16 in the [Interleukin-16]: Secreted and SUBCELLULAR LOCATION:. Tissue expression noted by UniProt (human): Expressed in hemopoietic tissues, such as resting T-cells, but undetectable during active T-cell proliferation. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
