Product Overview
This rabbit polyclonal antibody detects interleukin-18 (gene Il18) in mouse and rat samples and is validated for Western blot, flow cytometry and ELISA. The predicted molecular weight is 22.1 kDa, and the supplier reports an observed band at about 22 kDa.
Interleukin-18 (IL-18) is a pro-inflammatory IL-1 family cytokine made as an inactive precursor and cleaved by caspase-1 after inflammasome activation. Mature IL-18 signals through IL18R1 and IL18RAP to activate NF-κB and, together with IL-12, drives IFN-γ production by Th1 and NK cells.

Samples with a confirmed band (WB): rat thymus tissue, rat spleen tissue, rat lung tissue, rat liver tissue, mouse thymus tissue, mouse spleen tissue, mouse RAW264.7 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with a confirmed signal (flow cytometry): ANA-1 cells, mouse spleen tissue.

Recombinant mouse IL18 fragment (Asn36–Asn177), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with mouse and rat. UniProt describes interleukin-18 as secreted and also located in the cytoplasm. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
