Product Overview
This rabbit polyclonal antibody recognizes interleukin-33 (gene Il33) in mouse and rat samples and is validated for ELISA, IHC and Western blot. The predicted molecular weight is 30.0 kDa, and the supplier reports an observed band at about 35 kDa.
Interleukin-33 (IL-33) is an IL-1 family cytokine that acts as an “alarmin” released from damaged epithelial and endothelial cells. Binding to its receptor ST2 (IL1RL1) activates NF-κB and MAPK signaling, promoting type 2 immunity through Th2 cells, mast cells, eosinophils and group 2 innate lymphoid cells. Inside the nucleus, full-length IL-33 also associates with chromatin.

Samples with a confirmed band (WB): rat lung tissue, mouse lung tissue, mouse RAW264.7 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): mouse spleen tissue, rat lung tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).

Recombinant mouse IL33 fragment (Ala102–Ile266), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with mouse and rat. UniProt places interleukin-33 in the nucleus, chromosome, cytoplasm and cytoplasmic vesicle. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
