Product Overview
This rabbit polyclonal antibody targets inosine-5'-monophosphate dehydrogenase 2 (gene IMPDH2) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF and flow cytometry. The predicted molecular weight is 55.8 kDa, and the supplier reports an observed band at about 56 kDa.
IMPDH2 (inosine-5′-monophosphate dehydrogenase 2) catalyzes the conversion of IMP to XMP, the rate-limiting step in de novo guanine nucleotide synthesis. It is the predominant isoform in proliferating cells, including activated lymphocytes, and it is the target of immunosuppressive drugs such as mycophenolic acid. It is studied in cell growth, metabolism and cancer.

Samples with a confirmed band (WB): human K562 cells, human Daudi cells, human U251 cells, human U-937 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): T47D cells.
Samples with confirmed staining (ICC/IF): human breast cancer tissue.
Samples with a confirmed signal (flow cytometry): U2OS cells.
Immunogen
Synthetic peptide from the middle region of human IMPDH2. The peptide sequence is conserved in mouse and rat.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places inosine-5'-monophosphate dehydrogenase 2 in the cytoplasm and nucleus. Tissue expression noted by UniProt (human): IMPDH1 is the main species in normal leukocytes and IMPDH2 predominates over IMPDH1 in the tumor. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
