Product Overview
This rabbit polyclonal antibody detects phosphatidylinositol 3,4,5-trisphosphate 5-phosphatase 1 (gene INPP5D) in human samples and is validated for Western blot, flow cytometry and ELISA. The predicted molecular weight is 133.3 kDa, and the supplier reports an observed band at about 145 kDa.
SHIP1 (encoded by INPP5D) is an SH2 domain-containing inositol 5-phosphatase expressed mainly in blood cells. It converts PI(3,4,5)P3 to PI(3,4)P2, acting as a negative regulator of PI3K signaling downstream of immune receptors. SHIP1 limits activation of B cells, mast cells and myeloid cells and is studied in immunology and microglial biology.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.25 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with a confirmed signal (flow cytometry): THP-1 cells.
Immunogen
Recombinant human INPP5D fragment (Gln81–Glu1093), expressed in E. coli. Sequence identity with the mouse and rat orthologs: 87.6% and 87.5%, respectively.
Reactivity Notes
The supplier lists reactivity with human. UniProt places phosphatidylinositol 3,4,5-trisphosphate 5-phosphatase 1 in the cytoplasm, cell membrane, membrane raft and membrane. Tissue expression noted by UniProt (human): Specifically expressed in immune and hematopoietic cells. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
