Biological background
Protein function (datasheet): May mediate the control of various cellular processes by insulin. When phosphorylated by the insulin receptor binds specifically to various cellular proteins containing SH2 domains such as phosphatidylinositol 3-kinase p85 subunit or GRB2. Activates phosphatidylinositol 3-kinase when bound to the regulatory p85 subunit (By similarity). .
Scientific background (datasheet): Insulin receptor substrate 1 (IRS-1) is a signalling adapter protein that in humans is encoded by the IRS-1 gene. It is mapped to 2q36.3. This gene exhibited no intrinsic enzyme activity, and it can serve as a docking protein involved in binding and activating other signal transduction molecules after being phosphorylated on tyrosine by insulin receptor kinase. IRS1 plays a key role in transmitting signals from the insulin and insulin-like growth factor-1 (IGF-1) receptors to intracellular pathways PI3K/Akt and Erk MAP kinase pathways. IRS1 also plays important biological function for both metabolic and mitogenic (growth promoting) pathways. In addition to those, IRS1 is a key regulator of PI3K within malignant cells.
Cellular localization (datasheet): Cytoplasm. Nucleus. The mutant Huntingtin protein colocalizes with AKAP8L in the nuclear matrix of Huntington disease neurons. Shuttles between cytoplasm and nucleus in a Ran GTPase-independent manner.
Tissue details (datasheet): Expressed in osteoblasts, but not in osteoclasts. .
Sequence similarities (datasheet): Contains 1 IRS-type PTB domain.

- Supports comparative expression analysis across conditions, genotypes, or treatments when paired with appropriate controls.
- Useful for confirming target presence and subcellular distribution using orthogonal readouts (e.g., microscopy vs. immunoblotting).
Common research applications
- Western blot (WB): Compare relative target abundance and apparent size/isoforms across samples; interpret bands in light of expected MW and potential PTMs.
- ELISA: Measure target abundance in compatible matrices using a standard-curve readout; ensure dilution linearity and appropriate controls.
- Immunohistochemistry (IHC): Assess tissue distribution and cell-type patterns; interpret staining with appropriate negative controls and antigen context.
- Flow cytometry: Quantify target-positive populations in single-cell suspensions; pair with viability and isotype/FMO controls conceptually.
Notes for experimental interpretation
- Consider isoforms, post-translational modifications, and processing that can shift apparent molecular weight or localization.
- Cross-reactivity (datasheet): No cross-reactivity with other proteins
- Use appropriate positive and negative controls (e.g., KO/KD, blocking peptide, or isotype controls) to support specificity interpretation.
As a polyclonal antibody, this reagent may recognize multiple epitopes on the target, which can improve detection robustness but may require careful specificity controls.
