Product Overview

This rabbit polyclonal antibody detects integrin alpha-3 (gene ITGA3) in human and rat samples and is validated for Western blot, IHC, ICC/IF and flow cytometry. The predicted molecular weight is 116.6 kDa, and the supplier reports an observed band at about 155 kDa.

Integrin alpha-3 (ITGA3, CD49c) pairs with integrin beta-1 to form a receptor for laminins, especially laminin-332 and laminin-511. It is highly expressed in epithelial cells and supports their adhesion to basement membranes, and it is important for kidney, lung and skin integrity.

IHC (paraffin sections) 2–5 µg/mL (Human)
Immunocytochemistry / Immunofluorescence 5 µg/mL (Human)
Flow cytometry (fixed cells) 1–3 µg/1×106 cells (Human)

Samples with a confirmed band (WB): human U87 cells, human A431 cells, human A549 cells, human U2OS cells, rat C6 cells, rat PC-12 cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Samples with confirmed staining (IHC): human lung cancer tissue, human colorectal adenocarcinoma tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).

Samples with confirmed staining (ICC/IF): A431 cells.

Samples with a confirmed signal (flow cytometry): U87 cells.

Immunogen

Synthetic peptide from the N-terminal region of human integrin alpha 3. The peptide sequence is conserved in rat and mouse.

Reactivity Notes

The supplier lists reactivity with human and rat. UniProt places integrin alpha-3 in the cell membrane and cell projection. Tissue expression noted by UniProt (human): Isoform 1 is widely expressed. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

For research use only; not for diagnostic or therapeutic procedures. Handle with standard laboratory precautions.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.1–0.5 µg/mL (Human, Rat); IHC (paraffin sections) 2–5 µg/mL (Human); Immunocytochemistry / Immunofluorescence 5 µg/mL (Human); Flow cytometry (fixed cells) 1–3 µg/1×10⁶ cells (Human). Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Rat. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 116.6 kDa; observed band at about 155 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide from the N-terminal region of human integrin alpha 3. The peptide sequence is conserved in rat and mouse.
Q.How should this antibody be stored?
A.As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.
