Product Overview
This rabbit polyclonal antibody recognizes integrin alpha-4 (gene Itga4) in mouse and rat samples and is validated for Western blot, flow cytometry and ELISA. The predicted molecular weight is 115.7 kDa, and the supplier reports an observed band at about 150–180 kDa.
Integrin alpha-4 (ITGA4, CD49d) pairs with beta-1 to form VLA-4 and with beta-7 to form α4β7. These receptors bind VCAM-1, fibronectin and MAdCAM-1, guiding leukocyte adhesion to blood vessel walls and homing to inflamed tissue and the gut. Alpha-4 integrins are established targets in studies of leukocyte trafficking and inflammation.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Mouse, Rat) |
|---|---|
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Mouse) |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): rat liver tissue, mouse liver tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with a confirmed signal (flow cytometry): NIH/3T3 cells.
Immunogen
Recombinant mouse Integrin Alpha 4/Itga4 fragment (Tyr41–Asp963), expressed in E. coli. Sequence identity with the human ortholog: 85.2%.
Reactivity Notes
The supplier lists reactivity with mouse and rat. UniProt places integrin alpha-4 in the membrane. Tissue expression noted by UniProt (mouse): Expressed in the media layer of the arterial wall (at protein level). The samples tested by the supplier (listed above) are a practical starting point for positive controls.
