Product Overview
This rabbit polyclonal antibody recognizes integrin alpha-4 (gene ITGA4) in human samples and is validated for Western blot and ELISA. The predicted molecular weight is 114.9 kDa, and the supplier reports an observed band at about 140–150kDa.
Integrin alpha-4 (ITGA4, CD49d) pairs with beta-1 to form VLA-4 and with beta-7 to form α4β7. These receptors bind VCAM-1, fibronectin and MAdCAM-1, guiding leukocyte adhesion to blood vessel walls and homing to inflamed tissue and the gut. Alpha-4 integrins are established targets in studies of leukocyte trafficking and inflammation.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human) |
|---|---|
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): human Jurkat cells, human THP-1 cells, human HL-60 cells, human U87 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Immunogen
Recombinant human Integrin Alpha 4/ITGA4 fragment (Asn35–Val269), expressed in E. coli. Sequence identity with the mouse ortholog: 85.5%.
Reactivity Notes
The supplier lists reactivity with human. UniProt places integrin alpha-4 in the membrane. Tissue expression noted by UniProt (human): Expressed in vascular smooth muscle cells (at protein level). The samples tested by the supplier (listed above) are a practical starting point for positive controls.
