Product Overview
This rabbit polyclonal antibody detects integrin beta-1 (gene ITGB1) in human and rat samples and is validated for Western blot and IHC. The predicted molecular weight is 88.4 kDa, and the supplier reports an observed band at about 120–130 kDa.
Integrin beta-1 (ITGB1, CD29) is the most widely shared integrin beta subunit, pairing with many alpha subunits to form receptors for collagens, laminins, fibronectin and VCAM-1. Beta-1 integrins link the extracellular matrix to the actin cytoskeleton at focal adhesions and drive signaling for cell adhesion, migration, survival and tissue organization.

Validated Applications
| Western blot | 0.1–0.5 µg/mL (Human, Rat) |
|---|---|
| IHC (paraffin sections) | 0.5–1 µg/mL (Human) |
Samples with a confirmed band (WB): human A431 cells, rat C6 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human placenta tissue, human tonsil tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Immunogen
Recombinant human ITGB1 fragment (Asn527–Asp728), expressed in E. coli. Sequence identity with the mouse and rat orthologs: 91% and 88%, respectively.
Reactivity Notes
The supplier lists reactivity with human and rat. UniProt places integrin beta-1 in the cell membrane, cell projection, recycling endosome and melanosome. Tissue expression noted by UniProt (human): Expressed in vascular smooth muscle cells (at protein level)... The samples tested by the supplier (listed above) are a practical starting point for positive controls.
