Product Overview
This rabbit polyclonal antibody detects integrin beta-3 (gene ITGB3) in human, mouse and rat samples and is validated for Western blot and IHC. The predicted molecular weight is 87.1 kDa, and the supplier reports an observed band at about 29 kDa.
Integrin beta-3 (ITGB3, CD61) pairs with alpha-IIb to form the platelet fibrinogen receptor and with alpha-V to form the vitronectin receptor αVβ3. These integrins mediate platelet aggregation, cell adhesion to RGD-containing matrix proteins, angiogenesis and bone resorption by osteoclasts. Mutations in ITGB3 cause Glanzmann thrombasthenia.

Validated Applications
| Western blot | 0.1–0.5 µg/mL (Human, Mouse, Rat) |
|---|---|
| IHC (paraffin sections) | 0.5–1 µg/mL (Human, Rat) |
Samples with a confirmed band (WB): human U87 cells, human HEL cells, human HeLa cells, rat spleen tissue, mouse spleen tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human laryngeal squamous cells carcinoma tissue, human liver cancer tissue, human thyroid cancer tissue, rat lung tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Immunogen
Synthetic peptide from the C-terminal region of human integrin beta 3. The peptide sequence is conserved in rat and mouse.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places integrin beta-3 in the cell membrane, cell projection, cell junction and postsynaptic cell membrane. Tissue expression noted by UniProt (human): Isoform beta-3A and isoform beta-3C are widely expressed. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
