Product Overview
Clone AAFE-9 is a rabbit monoclonal antibody that recognizes integrin beta-4 (gene ITGB4) in human and mouse samples and is validated for Western blot and IHC. The predicted molecular weight is 202.2 kDa, and the supplier reports an observed band at about 240 kDa.
Integrin beta-4 (ITGB4, CD104) pairs with integrin alpha-6 to form a laminin receptor that is the core of hemidesmosomes. Its long cytoplasmic tail links to keratin filaments through plectin, anchoring epithelial cells to the basement membrane, and mutations in ITGB4 cause junctional epidermolysis bullosa with pyloric atresia.

Samples with a confirmed band (WB): human PC-3 cells, human Caco-2 cells, human HeLa cells, human MCF-7 whole cel.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 8% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human skin cancer tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).

Reactivity Notes
The supplier lists reactivity with human and mouse. UniProt places integrin beta-4 in the cell membrane and cell junction. Tissue expression noted by UniProt (human): Integrin alpha-6/beta-4 is predominantly expressed by epithelia. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
