Product Overview
This rabbit polyclonal antibody detects interferon regulatory factor 8 (gene IRF8) in human and mouse samples and is validated for Western blot, IHC, ICC/IF and flow cytometry. The predicted molecular weight is 48.4 kDa, and the supplier reports an observed band at about 48 kDa.
Interferon regulatory factor 8 (IRF8, also ICSBP) is a transcription factor expressed mainly in myeloid and lymphoid cells. It controls the development of monocytes, macrophages and dendritic cells, including conventional type 1 and plasmacytoid dendritic cells, and it regulates interferon-responsive genes in immune cells.

Validated Applications
| Western blot | 0.1–0.5 µg/mL (Human, Mouse) |
|---|---|
| IHC (paraffin sections) | 2–5 µg/mL (Human, Mouse) |
| Immunocytochemistry / Immunofluorescence | 5 µg/mL (Human) |
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human) |
Samples with a confirmed band (WB): human THP-1 cells, mouse spleen tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human tonsil tissue, mouse spleen tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): U2OS cells.
Immunogen
Synthetic peptide from the C-terminal region of human IRF8. It differs from the mouse and rat sequences at two residues.
Reactivity Notes
The supplier lists reactivity with human and mouse. UniProt places interferon regulatory factor 8 in the nucleus and cytoplasm. Tissue expression noted by UniProt (human): Predominantly expressed in lymphoid tissues. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
