Product Overview

This rabbit polyclonal antibody recognizes interleukin-6 (gene IL6) in human samples and is validated for Western blot. The predicted molecular weight is 23.7 kDa, and the supplier reports an observed band at about 20 kDa.

Interleukin-6 (IL-6) is a multifunctional cytokine released by macrophages, dendritic cells and many other cells during infection and tissue injury. It binds IL-6R and then the signaling subunit gp130, activating JAK–STAT3 through classic or trans-signaling. IL-6 drives the acute phase response, B-cell differentiation and Th17 development, and it is one of the most widely measured inflammatory markers.

Storage As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.

Validated Applications

Western blot 0.1–0.5 µg/mL (Human)

Conditions in the example images (WB): 5–20% gradient SDS-PAGE, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Immunogen

Recombinant human IL-6 fragment (Pro29–Met212), expressed in E. coli.

Reactivity Notes

The supplier lists reactivity with human. UniProt describes interleukin-6 as a secreted protein. Tissue expression noted by UniProt (human): Produced by skeletal muscle. A carrier-free (BSA-free) format can be requested; specify it in the order note.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.1–0.5 µg/mL (Human). Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human. Other species are not listed by the supplier.
Q.Has this antibody been tested on horse samples?
A.Reactivity with horse samples has not been tested by the supplier. Aligning the immunogen sequence with the horse ortholog (e.g., by BLAST) helps estimate the chance of cross-reactivity.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 23.7 kDa; observed band at about 20 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Recombinant human IL-6 fragment (Pro29–Met212), expressed in E. coli.
Q.How should this antibody be stored?
A.As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.
