Product Overview
This rabbit polyclonal antibody recognizes intersectin-1 (gene ITSN1) in human samples and is validated for Western blot, IHC, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 195.4 kDa, and the supplier reports an observed band at about 150 kDa.
Intersectin-1 (ITSN1) is a large adaptor protein with EH and SH3 domains that links endocytosis to the actin cytoskeleton. It helps assemble clathrin-coated pits by binding dynamin and other endocytic proteins, and its long isoform acts as a guanine nucleotide exchange factor for CDC42 to promote actin assembly.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human) |
|---|---|
| IHC (paraffin sections) | 2–5 µg/mL (Human) |
| Immunocytochemistry / Immunofluorescence | 5 µg/mL (Human) |
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human) |

Samples with a confirmed band (WB): human Caco-2 cells, human SiHa cells, human HeLa cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human brain tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): A549 cells.
Samples with a confirmed signal (flow cytometry): SiHa cells.
Immunogen
Recombinant human Intersectin 1/ITSN1 fragment (Ala522–Pro1721), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human. UniProt places intersectin-1 in the endomembrane system, synapse, cell projection and cell membrane. Tissue expression noted by UniProt (human): Isoform 1 is expressed almost exclusively in the brain. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
