Biological background

Protein function (datasheet): Chemotactic for monocytes and T-lymphocytes. Binds to CXCR3.

Scientific background (datasheet): C-X-C motif chemokine 10 (CXCL10), also known as Interferon gamma-induced protein 10 kDa (IP-10) or small-inducible cytokine B10, is a protein that in humans is encoded by the CXCL10 gene. It is a small cytokine belonging to the CXC chemokine family. The gene for CXCL10 is located on human chromosome 4 in a cluster among several other CXC chemokines. Luster et al. (1987) reported the isolation of an interferon-inducible gene that encodes a 98-amino acid protein called IP10. The chemokine IFN-gamma-inducible protein 10 (IP-10; CXCL10), a CXC chemokine, and its receptor, CXCR3, were found to be overexpressed in lymph node cells of EAMG rats. IP10 inhibits bone marrow colony formation, has antitumor activity in vivo, is a chemoattractant for human monocytes and T cells, and promotes T cell adhesion to endothelial cells.

Cellular localization (datasheet): Secreted.

Tissue details (datasheet): Expressed on activated B-cells.

Sequence similarities (datasheet): Belongs to the intercrine alpha (chemokine CxC) family.

Research relevance and current trends

  • Commonly studied in contexts related to Angiogenesis,Cancer,Chemokines,Immunology,Innate Immunity,Invasion/Microenvironment.
  • Supports comparative expression analysis across conditions, genotypes, or treatments when paired with appropriate controls.
  • Useful for confirming target presence and subcellular distribution using orthogonal readouts (e.g., microscopy vs. immunoblotting).

Common research applications

  • Western blot (WB): Compare relative target abundance and apparent size/isoforms across samples; interpret bands in light of expected MW and potential PTMs.
  • ELISA: Measure target abundance in compatible matrices using a standard-curve readout; ensure dilution linearity and appropriate controls.
  • Immunohistochemistry (IHC): Assess tissue distribution and cell-type patterns; interpret staining with appropriate negative controls and antigen context.
  • Flow cytometry: Quantify target-positive populations in single-cell suspensions; pair with viability and isotype/FMO controls conceptually.

Notes for experimental interpretation

  • Consider isoforms, post-translational modifications, and processing that can shift apparent molecular weight or localization.
  • Cross-reactivity (datasheet): No cross-reactivity with other proteins
  • Use appropriate positive and negative controls (e.g., KO/KD, blocking peptide, or isotype controls) to support specificity interpretation.

As a polyclonal antibody, this reagent may recognize multiple epitopes on the target, which can improve detection robustness but may require careful specificity controls.

Our team were content with the WB result of your anti-IP10/CXCL10 antibody. However we have seen positive staining in prostate secreted. using this antibody. Is that expected? Could you tell me where is CXCL10 supposed to be expressed?
According to literature, prostate does express CXCL10. Generally CXCL10 expresses in secreted. Regarding which tissues have CXCL10 expression, here are a few articles citing expression in various tissues: Foreskin keratinocyte, Pubmed ID: 11559369 Keratinocyte, Pubmed ID: 10233762 Prostate, Pubmed ID: 15489334
