Product Overview
This rabbit polyclonal antibody detects interferon regulatory factor 3 (gene IRF3) in human and mouse samples and is validated for Western blot, IHC, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 47.2 kDa, and the supplier reports an observed band at about 55 kDa.
Interferon regulatory factor 3 (IRF3) is a transcription factor that drives type I interferon production during viral infection. Sensing pathways such as RIG-I/MAVS, cGAS–STING and TLR3/4–TRIF activate the kinase TBK1, which phosphorylates IRF3 so it can dimerize, enter the nucleus and induce IFN-β and interferon-stimulated genes.

Samples with a confirmed band (WB): human HeLa cells, human Raji cells, human HepG2 cells, human A549- WT cells, human A549-IRF3 KO cells, mouse lung tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human rectal cancer tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): A431 cells.
Samples with a confirmed signal (flow cytometry): K562 cells.
Immunogen
Recombinant human IRF3 fragment (Met1–Ser427), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human and mouse. UniProt places interferon regulatory factor 3 in the cytoplasm, nucleus and mitochondrion. Tissue expression noted by UniProt (human): Expressed constitutively in a variety of tissues. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
